Epigenetic Reprogramming in Embryogenesis and the Germline Group, Gene Regulation, Stem Cells and Cancer Programme, CRG
Female mammals inactivate one of two X-chromosomes to achieve equal gene dosage with males, which have only one X. This X-inactivation process includes multiple epigenetic mechanisms and is closely linked to the differentiated state. X-inactivation is reversed in pluripotent stem cells, during development and in the germ cell lineage by X-chromosome reactivation, which renders both X-chromosomes active.
Here I will present different candidate and unbiased screening approaches, which our laboratory undertakes to elucidate the mechanisms of the X-chromosome reactivation process. We study epigenetic reprogramming of the X-chromosome in mouse embryos, during induced pluripotent stem cell (iPSC) reprogramming and during formation of the germ cell lineage. Thereby we want to gain further insights into how X-reactivation is linked to its biological context and how the erasure of epigenetic modifications and gene reactivation is accomplished.
